No. Spotify tracks now play right in the browser through an embedded player, same as YouTube and SoundCloud, so nobody needs an account or the app installed to join a Spotify room. Log into Spotify in that browser and you get the full track instead of a 30 second preview, but it is not required to play or vote.

To vote, no: viewers just type a number 0.5–5 in chat and it counts instantly. Submitting a song can work the same way (pasting a link in chat) or, if the host requires it, through a submit link viewers log into with their Twitch account.
